Xác định điều kiện nuôi cấy thích hợp để Lactobacillus plantarum W5 sinh exopolysaccharide cao.
Abstract
Ảnh hưởng của các điều kiện nuôi cấy đến khả năng sinh exopolysaccharide (EPS) của vi khuẩn Lactobacillus plantarum W5 (L. plantarum W5) được phân lập từ dịch whey sữa đậu nành đã được khảo sát. Kết quả cho thấy khi nuôi L. plantarum W5 trong môi trường MRS có bổ sung nguồn C và N sẽ làm tăng đáng kể khả năng sinh EPS của nó. Trong 3 loại đường khảo sát, saccharose với nồng độ bổ sung 4% giúp thu được lượng EPS cao nhất là 167,232 µg/ml. Và trong môi trường có saccharose thì 0,4% cao nấm là nguồn N thích hợp cho L. plantarum W5 sinh EPS cao với lượng thu được là 304,427µg/ml. Kết quả nghiên cứu tiếp theo cho thấy các điều kiện nuôi cấy khác cũng có ảnh hưởng đáng kể đến khả năng sinh tổng hợp EPS của L. plantarum W5. Khi nuôi cấy L. plantarum W5 trong môi trường có bổ sung nguồn C và N thích hợp thì mật độ tế bào gieo cấy ban đầu 106 CFU/ml là tốt nhất với hàm lượng EPS thu được là 306,459 µg/ml. pH môi trường ban đầu bằng 6, nhiệt độ nuôi cấy 35oC và thời gian nuôi cấy 36h là điều kiện nuôi cấy tốt nhất để L. plantarum W5 sinh tổng EPS với hàm lượng thu được 447,598 µg/ml.References
